Peptide Therapeutics

Peptide therapeutics are short chains of amino acids (typically 2-50) that act as drugs, signaling molecules, and delivery vehicles. The field has been revolutionized by GLP-1 receptor agonists like semaglutide (Ozempic/Wegovy) and tirzepatide (Mounjaro/Zepbound), which have become the best-selling drug class in history for treating obesity and diabetes. Beyond metabolic medicine, peptides play a growing role in longevity science (MOTS-c, GHK-Cu, Epitalon), CRISPR delivery via cell-penetrating peptides, and peptide-based partial epigenetic reprogramming, making them a bridge between traditional pharmacology and genetic medicine.

Key Glossary Terms

amino acid

The building block of peptides and proteins. There are 20 standard amino acids, each with a unique side chain. They link via peptide bonds to form peptide chains.

bioavailability

The fraction of an administered drug that reaches systemic circulation. Peptides historically have very low oral bioavailability due to gut proteases and poor membrane permeability — which is why most are injected.

BPC-157

Body Protection Compound 157 — a synthetic pentadecapeptide derived from a gastric protein. Popular in biohacker circles for claimed tissue-repair effects, but lacks human clinical trials, is banned by WADA (2022), and is restricted by FDA 503A compounding rules (2023). Not FDA-approved.

cell-penetrating peptide

A short peptide (typically 5–30 amino acids, often cationic like poly-arginine or TAT) that can cross cell membranes and carry cargo such as proteins, nucleic acids, or CRISPR ribonucleoproteins into cells. A promising non-viral delivery method.

CJC-1295

A synthetic analog of growth hormone-releasing hormone (GHRH) designed for long half-life via albumin binding. Sold as a research chemical; not FDA-approved.

CPP

Cell-penetrating peptide. See full entry.

cyclic peptide

A peptide whose ends are joined to form a ring, or whose side chains are linked to create a macrocycle. Cyclization improves stability, protease resistance, and target affinity. Examples include cyclosporine and oxytocin.

Epitalon

A synthetic tetrapeptide (Ala-Glu-Asp-Gly) developed by Vladimir Khavinson in Russia. Claimed to activate telomerase and extend lifespan based primarily on Russian rodent studies and small human trials. Not FDA-approved; sold as a research chemical.

GHK-Cu

A copper-binding tripeptide (Gly-His-Lys + Cu²⁺) discovered by Loren Pickart in 1973. Modulates expression of roughly 4,000 genes and is used in cosmetics for skin repair. Injectable use is a regulatory gray zone.

GIP

Glucose-dependent insulinotropic polypeptide — an incretin hormone released from intestinal K-cells. Tirzepatide activates both GIP and GLP-1 receptors for synergistic weight loss effects.

GLP-1

Glucagon-like peptide-1, a natural incretin hormone released from gut L-cells after meals. It stimulates insulin release, suppresses glucagon, slows gastric emptying, and reduces appetite. GLP-1 receptor agonists like semaglutide and tirzepatide are now blockbuster obesity and diabetes drugs.

GLP-1 receptor agonist

A drug that binds and activates the GLP-1 receptor, mimicking the natural hormone to treat type 2 diabetes and obesity. Examples include semaglutide (Ozempic, Wegovy), liraglutide, and dulaglutide.

incretin

A class of gut hormones (primarily GLP-1 and GIP) released after eating that stimulate insulin secretion in a glucose-dependent manner. Incretin-based therapies are the foundation of modern obesity and diabetes drugs.

Ipamorelin

A selective growth hormone secretagogue peptide that stimulates GH release without affecting cortisol or prolactin. Sold as a research chemical; not FDA-approved.

lipidation

The covalent attachment of a fatty acid chain to a peptide to enable albumin binding in the bloodstream, dramatically extending half-life. Used in semaglutide and tirzepatide.
